Abstract The aqueous two-phase system (ATPS) as an environmentally friendly technique has a high potential for extraction of drugs, proteins, metals, etc. In this work first the formation ATPSs by betaine, which is novel, low toxic, and biodegradable material, polyethylene glycol or phosphate salts (K 3 PO 4 K 2 HPO ) were investigated temperatures 298.15, 308.15, 318.15 atmospheric pressure (≈ 85 kPa). effect increasing temperature on binodal curves was investigated. experimental data correlated using two empirical nonlinear three parameter expressions developed Merchuk, Zafarani-Moattar et al. two-parameter modified equation derived from effective excluded volume theory. The separation of large polar constituents presents a substantial challenge in natural product research when employing column chromatography techniques, as the process is both complex and time-consuming. In this study, an acetonitrile/tetrahydrofuran/di-(2-ethylhexyl) phosphoric acid/aqueous saturated sodium chloride solvent system was developed utilized for countercurrent from Ginkgo biloba L. seeds. Five were effectively isolated using (2:2:0.8:3, v/v) two-step method. initial process, three successfully purified methanol extract: compound 1, 4, 5. Compounds 2 3, co-eluted column, further subjected to inner-recycling chromatographic procedures. At last, five identified, including 4′-O-methylpyridoxine (1); two indole alkaloid N-glucosides, ginkgoside B (2) A (3); 2-(4-hydroxybenzyl) malic acid (4); coniferyl alcohol (5). results demonstrated that serves feasible efficient components.

These successfully separated at purities 72.88%, 98.55%, 96.86%, 96.74%, 95.84%, respectively, using n ‐hexane/ethyl acetate/methanol/water (4.0/5.0/4.0/5.0, v/v/v/v), (5.0/5.0/6.0/4.0, (4.9/5.1/5.7/4.3, v/v/v/v) mobile phase systems. revealed that E had best inhibitory effect.
