Ulcerative colitis, a chronic inflammatory condition of the colon, requires precise and targeted treatment, polysaccharides, with their pH responsiveness biodegradability, offer an innovative approach for colon-specific drug delivery. This study aims to develop highly delivery system enhanced therapeutic targeting efficiency ulcerative focusing on preparation, optimisation, evaluation dual cross-linked mesalamine-loaded sericin-pectin (DCLSPs) micro-beads. These beads utilise pH-responsive microflora biodegradability properties polysaccharides colon delivery, employing Response Surface Methodology. Formulated via ionotropic gelation method divalent cross-linking ions (Ca2+ SO42–), DCLSPs were optimised using Box-Behnken design assess impact varying drug, pectin, sericin polymer proportions. The evaluated entrapment efficiency, thermal behaviour, surface morphology, water uptake, swelling, in-vitro release. Results indicated that spherical successfully developed, encapsulation ranging from 65.1% 95.5%, loading between 32.5% 49.9%, bead sizes 0.75 mm 0.92 mm, degrees swelling 1.82.
