Nattokinase (NK), a serine protease with high thrombolytic activity, has significant potential for application in foods intended special health benefits. However, the NK production wild-type Bacillus subtilis natto is relatively low. In this study, high-yielding and genetically stable mutant strain (B. JNC002.001, 300.0 ± 4.7 FU/mL) was obtained through atmospheric room temperature plasma (ARTP) mutagenesis. It increased activity by 1.84 times compared to initial SD2, demonstrating prospects food fermentation applications. Additionally, B. JNC002.001 exhibited notable alterations growth characteristics, glucose consumption, sporulation. This study further elucidated mechanism of enhanced at molecular level. Genome resequencing revealed that genes included 10 single nucleotide polymorphisms (SNPs) one insertion, among which kinA gltA were associated sporulation synthesis, respectively. terms transcriptional level, NK-coding gene aprN up-regulated 9.4 relative strain. Most related central carbon metabolism Sec secretion pathway up-regulated. addition, expression regulatory factors transcription process provided evidence deficiency JNC002.001. These results could provide insights into facilitate construction engineered strains yield.
