Invertases (INVs) included CWIN, CIN and VIN, are key enzymes in sucrose hydrolysis into glucose fructose essential for plant root development. Yet the effects of nitrogen planting density on IbINVs expression remains unexplored sweetpotato. This study identified 22 invertase (IbINV) genes sweetpotato genome conducted comprehensive analyses their subcellular localization, gene structure, conserved motifs domains. Gene Ontology functional protein interaction network analysis suggested that IbCWIN1/2/5/6 potentially interact with HKL1/3, HXK2/3/4, other proteins, significantly influencing carbohydrate metabolic functions biological processes Transcriptome data revealed IbCIN2 IbVIN3 were highly expressed fibrous (FR) potential storage (PSR), while IbVIN4 exhibited high levels roots (SR), IbCWIN2 was both FR SR. Moreover, field experiments demonstrated that, compared EN (180 kg N ha-1) combined LD (50,000 ha− 1), MN (120 1) MD (62,500 enhanced number weight. Notably, LDEN treatment, IbCIN2, IbCWIN2, under MDMN treatment upregulated, extremely significant differences at 15DAP, which showed a maximum 23.24-fold change, showing positive correlation increased INV enzyme activity, (GLc), (FRc) content. Additionally, screening homologous is to AtA/N-INVI AtA/N-INVG, VACUOLAR, INVERTASE1 BFRUCT4, similar functions, further supporting involvement formation These observations underscore expansion provide theoretical basis optimizing between application sweetpotato, allowing more efficient use improved growth.
