To investigate the structure-antioxidant activity relationship, Pleurotus ferulae polysaccharides were extracted using ultrasonic (U-PFPS) and microwave/ultrasonic-assisted methods (MU-PFPS). Compared to U-PFPS with a molecular weight of 1.566 × 103 kDa, MU-PFPS exhibited lower 89.26 kDa. In addition, unlike U-PFPS, which is primarily composed glucose (Glu:Man:Gal = 91.1:3.5:5.4), has more balanced composition Glu:Man:Gal in ratio 39.4:27.8:32.8 contains branched chains. Furthermore, antioxidant analysis revealed that high concentration (at concentrations above 600 μg/mL) demonstrated stronger protective effects against oxidative damage RAW264.7 cells than did. Collectively, these data suggest higher branching degree at appropriate may correlate enhanced enzyme activities. Our work provides method for isolating offers insights into structure-activity relationship polysaccharides, laying foundation future applications polysaccharide modification structural characterization.

ABSTRACT Type 2 diabetes mellitus (T2DM), a widespread chronic metabolic disorder, presents global challenge. Metformin hydrochloride, although widely prescribed, is associated with notable side effects. This study aims to explore safer, more effective alternatives by assessing the impact of Ruditapes philippinarum polysaccharides (RPPs) on glycemic control and modulation microbiota in T2DM mice. A mouse model was established through high‐sucrose/high‐fat diet intraperitioneal streptozotocin injection. Glycometabolism indicators, histopathological features, gut composition caecum samples were assessed. Following 4 weeks RPPs intervention, fasting blood glucose (FBG), glycated serum protein (GSP), area under curve (AUC) oral tolerance test (OGTT), total cholesterol (TC), triglyceride (TG), low‐density lipoprotein (LDL‐c) levels reduced mice, while AKT‐1 GLUT‐2 transcription significant increased. Short‐chain fatty acids (SCFAs) concentrations notably increased RPP‐L group compared Model group, improvements also observed medium‐chain (MCFAs) secondary bile (SBAs). Regarding microbiota, Firmicutes‐to‐Bacteroidetes (F/B) ratio substantially lower than group. At genus level, beneficial bacteria, such as Akkermansia , Alloprevotella Tidjanibacter Faecalibaculum demonstrated abundance. Correlation analysis identified Muribaculum Paramuribaculum Lacrimispora Turicibacter microbial taxa progression. significantly alleviated hyperglycemic symptoms mice enhancing presence leading marked improvement intestinal composition. research offers foundational insights for potential use R. biomedical nutraceutical applications.
