DNA technology has emerged as a promising route to accelerated manufacture of sequence agnostic vaccines. For activity, vaccines must be protected and delivered the correct antigen presenting cells. However, physicochemical properties vector carefully tuned enhance interaction with immune cells generate sufficient response for disease protection. In this study, we have engineered range polymer-based nanocarriers based on poly(beta-amino ester) (PBAE) polycation platform investigate role that surface poly(ethylene glycol) (PEG) density pDNA encapsulation, formulation gene transfectability both in vitro vivo. We achieved by synthesising non-PEGylated PEGylated PBAE produced formulations containing these PBAEs, mixed polyplexes tune PEG density. Despite high transfection HEK293T cells, only fully displayed significantly higher expression reporter than negative control dendritic Further vivo studies bivalent SARS-CoV-2 vaccine revealed led strong specific T-cell responses, however did not translate into presence serum antibodies indicating need further improving immunisation polymer delivery systems.
