Despite its advances in medicine, cancer still remains elusive for most of the conventional therapies because medications are applied to healthy tissues as well; traditional procedures frequently result significant adverse effects. This chapter provides a comprehensive overview role nanotechnology diagnosis, imaging, and treatment. The explores both passive active targeting techniques well mechanics behind medication delivery using nanoparticles. Therefore, potential improve biomarker detection imaging is investigated. Furthermore, prospect co-delivery numerous therapeutic drugs into single nanocarrier combination therapy investigated, emphasising synergistic benefits improved results possible with this strategy. Overall, emphasises how much may do enhance outcomes by offering focused, effective, least intrusive treatment options.

Lipid nanoparticles (LNPs) have been utilized to deliver small interfering RNA (siRNA) treat acute liver injury. However, LNPs exhibit suboptimal lysosomal escape capabilities and biodegradability. To address these limitations, we designed triglyceride-mimetic ionizable lipids by conjugating N,N-dimethyl tertiary amine head groups the sn-2 position of triglyceride (TG) through ester bonds. These were abbreviated as 2C-TG, 3C-TG, 4C-TG, with located in β-, γ-, δ-positions linkage bond, respectively. The uniform-size prepared using ethanol dilution method. Notably, group within carbon chain is found significantly influence critical parameters, including encapsulation rate, pKa, cellular uptake, escape, lipase release, gene silencing efficiency. Our findings hold promise for improving efficacy safety LNP-based siRNA therapeutics.
