ABSTRACT Background Chronic rhinosinusitis (CRS) is characterized by persistent nasal and paranasal sinus mucosa inflammation. It comprises two phenotypes, namely CRS with polyps (CRSwNP) without (CRSsNP). CRSwNP can be associated asthma hypersensitivity to non-steroidal anti-inflammatory drugs (NSAIDs) in a syndrome known as NSAID-exacerbated respiratory disease (N-ERD). Furthermore, frequently intertwines allergies. Objective This study investigated the phenotypic characteristics of peripheral blood mononuclear cells (PBMCs) within cohorts patients, additionally examining influence comorbid allergies on these parameters. Methods 24 participants were grouped into controls, CRSsNP, CRSwNP, N-ERD ( n=6 /group), half patients each group having Levels cytokines quantified secretions sera. The abundance features immune PBMCs evaluated through mass cytometry clustering methods. Results showed heightened type 2 cytokine levels. Mass analysis revealed increased activated naive B cell levels N-ERD, while resting higher CRSsNP. Th2a did not differ between subtypes but significantly elevated allergic subjects. In had lower CXCR5 CD45RA expression, NK displayed reduced CD56 Conclusions There are distinct immunological phenotypes allergy, Capsule summary examines profiles different allergy highlighting intricacies differences allergy.

BackgroundA preference for type 2 immunity plays a central role in the pathogenesis of atopic dermatitis (AD). Dupilumab, an mAb targeting IL-4 receptor α (IL-4Rα) subunit, inhibits and IL-13 signaling. These cytokines contribute significantly to IgE class switch recombination B cells, critical diseases. Recent studies indicate IgG+CD23hiIL-4Rα+ memory cells (MBC2s) as IgE-producing B-cell precursors, linked total serum levels patients. Total decreased during dupilumab treatment previous studies.ObjectiveWe sought assess effects comparison with alternative therapies on frequency MBC2s correlation pediatric patients AD.MethodsPediatric AD, participating ongoing trial, underwent randomization into 3 groups: (n = 12), cyclosporine topical 12). Plasma samples PBMCs were collected at baseline (T0) 6 months after starting therapy (T6). Flow cytometry was used PBMC phenotyping, ELISA plasma.ResultsOur findings revealed significant reduction MBC2 among treated dupilumab. In addition, observed between levels.ConclusionsSystemic blocking IL-4Rα subunit leads decrease circulating AD. Our unveiled novel mechanism through which exerts its influence signature.

Five recombinant fusion proteins (PreS-Cat 1-PreS-Cat 5) each containing hepatitis B virus (HBV)-derived PreS as an immunological carrier non-allergenic peptides from sites expressed in Escherichia coli, purified, characterized mass spectrometry, circular dichroism (CD), size exclusion chromatography. Asthma is a complex, multifactorial inflammatory disorder of the airways, characterized by recurrent symptoms and variable airflow obstruction. So far, two main asthma endotypes have been identified, type 2 (T2)-high or T2-low, based on underlying immunological mechanisms. Recently, extracellular vesicles (EVs), particularly exosomes, gained increasing attention due to their pivotal role in intercellular communication distal signaling modulation. In context pathobiology, an amount experimental evidence suggests that EVs secreted eosinophils, mast cells, dendritic T neutrophils, macrophages, epithelial cells contribute disease This review explores profiling molecular signatures T2-high T2-low asthma, offering novel perspectives mechanisms potential therapeutic targets.

Abstract Studies about thymic B cells are scarce in the literature, but it was suggested that they can exert modulatory and regulatory functions on immune system. Thymic play some role regulating most frequent allergic background worldwide, atopy induced by mite Dermatophagoides pteronyssinus (Der p). Here, we aimed to evaluate if polyclonal IgG repertoire produced Der p-atopic individuals influence homing cytokine profile of human derived from non-atopic children aged less than seven days.
