Introduction Food allergies, like cow’s milk allergy, significantly impact children, with sensitization often beginning during the first year of life. Human oligosaccharides (HMOs) may influence this process, as specific HMOs differentially affect mucosal immune responses in vitro . Given distinct HMO profiles secretor (Se+) and non-secretor (Se-) milk, we investigate how full from Se+ Se- absence or presence a allergen. Methods Monocyte-derived dendritic cells (moDCs) were exposed to isolated pooled (pHMOs), subsequently co-cultured naïve T confirm modulation. We compared type 2-activation capability several proteins via direct exposure moDCs intestinal epithelial (IECs) moDCs. Finally, studied effect pHMOs allergen β-lactoglobulin (BLG) (via (IECs)) on subsequent cell response. Results Both dose-dependently activated moDCs, indicated by increased IL8 release %CD80+ tended increase 2-associated markers, while also increasing regulatory IL10 release. pHMOs-pre-exposed instructed produce 2 cytokines IL13. reduced %CD86+ but did not drive signature cells. In BLG, IL13 cells, percentage contrast, co-exposure BLG only slightly affected moDC phenotype, these modify phenotypes. Conclusions activation. pHMO-pre-exposed induced 2- regulatory-associated phenotype. These data suggest that depending status, modulate responsiveness
