In this study, an ultrasonic-assisted procedure for the extraction of mulberry leaf polysaccharides (MLPs) was investigated using response surface methodology with a 29-run Box–Behnken design. Four factors were investigated, and it found that influencing process, in order significance, temperature > liquid-to-material ratio ultrasonic power. Considering practical conditions, parameters adjusted to 16:1 mL/g, time 58 min, 65 °C, power 500 W. Under these yield MLPs 14.47%, which is close predicted value, indicating process optimized by (RSM) feasible. The separation purification effects macroporous resin activated carbon on D152 being considered most suitable choice. optimal conditions be sample concentration 0.5 g/mL flow rate 1 mL/min. Thin-layer chromatography infrared spectroscopy revealed extracted from leaves are primarily composed rhamnose, xylose, arabinose. conclusion, study successfully through methodology, determined parameter combination, verified its efficiency stability. obtained extraction, significantly higher than reported previous literature. decolorization crude polysaccharide extract also purified have clear monosaccharide composition structural characteristics, providing theoretical basis technical support their application functional food or drug development.
