Genes discovered by previous epigenetic studies of schizophrenia have focused solely on diagnostics or pathology, potentially leading to a disconnection between them. Using these molecules identify the disease is considered insufficient. MicroRNAs (miRNAs) binding messenger RNAs (mRNAs) can lead mRNA degradation, while circular (circRNAs), miRNAs as sponge, reduce inhibitory effect mRNAs. CircRNAs, miRNAs, and mRNAs form multi-molecular axis that bind regulate expression each other, thereby affecting biological function. This study early-onset (EOS), aiming consisting circRNAs, with both neurobiological function diagnostic value assist in identification. With increasing use of medical imaging (e.g., CT scans) and environmental radiation sources, over 2 % pregnancies worldwide are inadvertently exposed to low-dose ionizing (IR), raising urgent concerns about fetal neuroprotection. While prenatal IR is implicated in microcephaly lifelong neuropsychiatric risks, prior studies have not resolved whether sirtuin-mediated pathways, particularly SIRT1/TPH2 signaling, drive these deficits or dietary phytochemicals like resveratrol can mitigate them. To determine (1) the role signaling IR-induced neurodevelopmental psychiatric impairments, (2) therapeutic potential maternal supplementation counteract effects-a strategy previously explored models. Mouse cohorts received X-ray irradiation (0, 1.0 Gy, Gy gestational day 8) with/without supplementation, followed by longitudinal cortical behavioral analyses. RNA sequencing/Western blotting quantified SIRT1, TPH2, BDNF, senescence markers (P16, P21 SA-β-gal). 5-HT levels were assessed ELISA. Depression-like behaviors tested via forced swim tail suspension. IR-exposed fetuses exhibited progressive with reduced thickness, accompanied SIRT1 downregulation, BDNF suppression, elevated cellular senescence. Adult offspring displayed depression-like behaviors, linked TPH2 downregulation diminished levels. Resveratrol normalized restored neurotrophic factors, attenuated both depressive phenotypes. This study provides first evidence that a central mediator prevents damage depression rescuing this pathway. These findings position as novel, mechanism-driven intervention for neuroprotection against radiation.
