BFSP1 (beaded filament structural protein 1) is a plasma membrane, Aquaporin 0 (AQP0/MIP)-associated intermediate expressed in the eye lens. myristoylated, post-translation modification that requires caspase cleavage at D433. Bioinformatic analyses suggested sequences 434-452 were α-helical and amphipathic.

Positioned within the eye, lens supports vision by transmitting and focusing light onto retina. As an adaptive glassy material, is constituted primarily densely-packed, polydisperse crystallin proteins that organize to resist aggregation crystallization at high volume fractions, yet details of how crystallins coordinate with one another template maintain this transparent microstructure remain unclear. The role individual subtypes (α, β, γ) paired subtype compositions, including they experience crowding-induced turbidity in solution, explored using combinations spectrophotometry, hard-sphere simulations, surface pressure measurements. After assaying combinations, β-crystallins emerged as a principal component all mixtures enabled dense fluid-like packing short-range order necessary for transparency. These findings helped inform design lens-like hydrogel systems, which are used monitor manipulate loss transparency under different crowding conditions. When taken together, illustrate characterization materials made from can be better understand mechanisms regulating

Ionising radiation (IR) is a cause of lipid peroxidation, and epidemiological data have revealed correlation between exposure to IR the development eye lens cataracts. Cataracts remain leading blindness around world. The plasma membranes fibre cells are one most cholesterolrich in human body, forming rafts contributing biophysical properties membrane. Liquid chromatography followed by mass spectrometry was used analyse bovine membrane fractions after 5 50 Gy lenses taken from wholebody 2 Gy-irradiated mice. Although cholesterol levels do not change significantly, dose-dependant formation oxysterols 7β-hydroxycholesterol, 7-ketocholesterol 5, 6-epoxycholesterol nucleus extracts observed. Whole-body X-ray (2 Gy) 12-week old mice resulted an increase 7β-hydroxycholesterol their lenses. Their regressed over 24 h living cortex exposure. This study also demonstrated that IR-induced fold greater mouse than nucleus. Further work required elucidate mechanistic link(s) cataract, but these evidence for first time results oxysterol

ABSTRACT Summary Statement This study illustrates a vital role for ankyrin-B in lens architecture, growth and function through its involvement membrane protein spectrin-actin cytoskeletal organization stability The transparent ocular is essential vision by focusing light onto the retina. Despite recognizing importance of unique cellular architecture mechanical properties, molecular mechanisms governing these attributes remain elusive. aims to elucidate (AnkB), scaffolding protein, cytoarchitecture, using conditional knockout (cKO) mouse model. AnkB cKO has no defects morphogenesis, but exhibited changes that supported global maintenance clarity, size, stiffness. Notably, absence led nuclear cataract formation, evident from P16. fibers exhibit progressive disruption cytoskeleton, channel proteins, cell-cell adhesion, shape change, loss degradation several proteins (e.g., NrCAM. N-cadherin aquaporin-0) along with disorganized plasma impaired ball-and-socket interdigitations. Furthermore, decreased Collectively, results illustrate organization.
