Scorpion is a commonly used drug in traditional Chinese medicine for treating epilepsy, although the exact mechanisms are not yet fully understood. This study aimed to compare treatment effects of water extract (SWE) and ethanol (SEE) on mice with pentetrazole-induced epilepsy investigate possible through metabolomics methods. A epileptic model was assess corrective SWE SEE. Untargeted metabolomics, utilizing UPLC-Q-TOF/MS, employed analyze metabolic profiles identify changes following scorpion treatment. The results revealed that only showed therapeutic mice. Metabolomics analysis demonstrated significant alterations signatures between groups. By orthogonal partial least squares discrimination analysis, 44 108 potential biomarkers mouse serum were identified positive negative ion modes, respectively. Differential metabolites related then pinpoint relevant pathways mice, such as linoleic acid metabolism, biosynthesis unsaturated fatty acids, glycerophospholipid ether lipid metabolism. In conclusion, this highlights provides insight into underlying involved its efficacy.

Plasma contains metabolites with diverse physicochemical properties, ranging from highly polar to apolar, and concentrations spanning at least nine orders of magnitude. metabolome analysis is valuable for monitoring health evaluating medical interventions but challenging due the metabolome’s diversity complexity. This study aims develop validate targeted LC-MS/MS methods quantifying 235 mammalian 17 compound classes in porcine plasma without prior derivatization. Utilizing reversed-phase hydrophilic interaction liquid chromatography coupled tandem mass spectrometry, each analyte identified quantified using two selected reaction (SRM) transitions. Fast polarity switching scheduled SRM enhance coverage throughput, enabling one sample about 40 min. A simple “dilute shoot” preparation protocol was employed, samples injected dilution levels align metabolite within calibration curve ranges. Validation included assessments carryover, linearity, detection quantification limits, repeatability recovery. The method further applied various animal species, demonstrating its applicability human studies. establishes robust comprehensive quantification, advancing large-scale metabolomics biomedical research.

Abstract High-performance liquid chromatography (HPLC) is a common medium-throughput technique to quantify the components of often complex mixtures like those typically obtained from biological tissue extracts. However, analysis HPLC data multianalyte samples hampered by lack tools accurately determine precise analyte quantities on level precision equivalent mass-spectrometry approaches. To address this problem, we developed tool call PeakClimber, that uses sum exponential Gaussian functions deconvolve overlapping, peaks in traces. Here analytically show are well-fit an function, PeakClimber more quantifies known peak areas than standard industry software for both and mass spectrometry applications, differences triglyceride abundances between colonized germ-free fruit flies.
