Scorpion is a commonly used drug in traditional Chinese medicine for treating epilepsy, although the exact mechanisms are not yet fully understood. This study aimed to compare treatment effects of water extract (SWE) and ethanol (SEE) on mice with pentetrazole-induced epilepsy investigate possible through metabolomics methods. Plasma contains metabolites with diverse physicochemical properties, ranging from highly polar to apolar, and concentrations spanning at least nine orders of magnitude. metabolome analysis is valuable for monitoring health evaluating medical interventions but challenging due the metabolome’s diversity complexity. This study aims develop validate targeted LC-MS/MS methods quantifying 235 mammalian 17 compound classes in porcine plasma without prior derivatization. Utilizing reversed-phase hydrophilic interaction liquid chromatography coupled tandem mass spectrometry, each analyte identified quantified using two selected reaction (SRM) transitions. Sinomenium acutum (Thunb.) Rehd. et Wils is widely used in the treatment of rheumatoid arthritis, with its alkaloid compound sinomenine (SIN) being renowned for significant anti-inflammatory properties. However, despite widespread application, vivo mechanisms and metabolic pathways SIN remain incompletely understood. This study established a rapid reliable method based on an ultra-high-performance liquid chromatography coupled Quadrupole-Exactive Orbitrap mass spectrometry molecular docking to identify characterize 69 metabolites rat plasma, urine, feces, revealing primary hydroxylation, demethylation, sulfation, glucuronidation. Molecular results revealed that phase I reactions, including dedimethylation, dehydrogenation, dihydroxylation, along their composite were pivotal influencing SIN's activity.
