Sealing and Chromatographic Performance of Microgroove Columns

In
a
recent
paper,
we
introduced
the
concept
of
structured
microgroove
columns
as
potential
solution
to
obtain
perfectly
ordered
particle
beds
for
nano-
and
micro-LC.
this
concept,
spherical
particles
are
either
individually
positioned
(single
layer
column)
or
stacked
(multilayer
in
series
interconnected
micromachined
pockets.
After
introducing
suitable
method
fill
efficiently
one-particle-per-pocket
mode,
present
contribution
addresses
issue
sealing
filled
reports
on
first
flow
tests
conducted
column
device.
For
purpose,
an
adapted
set
was
with
10
μm
calibrated
silica
subsequently
anodically
bonded
borofloat
top
plate.
The
chromatographic
performance
prototype
is
determined
by
injecting
band
fluorescent
dye
into
unretentive
column.
Reduced
plate
heights
were
higher
(hmin
=
2
instead
1)
than
theoretically
expected,
while
pressure
drop
resistance
much
closer
expected
value
(ϕi,i

versus

expected).
It
assumed
height
deviations
from
theory
due
fact
that
pockets
somewhat
oversized
(some
75%),
creating
unnecessary
large
dead
spaces.
