Messenger RNA (mRNA) has great potential to provide innovative medical solutions in the treatment of heart failure. Although lipid nanoparticles (LNPs) are an established mRNA delivery system, effectively delivering LNPs remains a significant challenge. Here, we evaluated efficacy transcatheter intracoronary (IC) administration compared intravenous (IV) and intramyocardial (IM) normal ischemia-reperfusion (I/R) model rabbit hearts using encapsulating Firefly Luciferase (FLuc) mRNA. In model, IVIS spectrum data showed that FLuc expression was widespread throughout IC group significantly higher than IV group, comparable IM where it highly expressed only at injection sites. Histological analysis revealed FLuc-expressing cells were observed cardiomyocytes, endothelial cells, smooth muscle fibroblasts. I/R also group. strongly infarcted area all three groups, demonstrated most remote area. other groups. delivered mRNA-LNPs not damaged but (non-damaged area) diseased heart. Moreover, VEGF mRNA-LNP via method reduced infarct attenuated impairment cardiac function caused by injury methods. Considering invasiveness clinically limited applications administration, our study suggests less invasive is safe useful for wider range myocardial tissue
