Reactive oxygen species (ROS)-sensitive polymers are extensively used in cancer therapies. However, the ROS levels tumor microenvironment often insufficient to trigger an adequate therapeutic response. Herein, we report a cinnamaldehyde (CA)-based ROS-responsive cationic polymer (PCA) and demonstrate its high efficiency gene delivery cell growth inhibition. CA could be released from via ROS-sensitive thioacetal bond by endogenous ROS. The successively induced more accumulation through GSH depletion, positive feedback helped PCA achieve self-accelerating degradation. Results proved that PCA/p53 complexes were efficient depleting GSH, upregulating levels, transfection. Besides, was also shown effective delivering p53. More importantly, significantly induce suppression synergistic effect of p53, providing valuable insights into design self-amplifying polymeric vectors.
