Cystobacter fuscus HM-E: a novel biocontrol agent against cotton Verticillium wilt DOI Creative Commons

Verticillium wilt of cotton, caused by dahliae , is one the most devastating soilborne fungal diseases in cotton production, urgently demanding development effective control measures. Myxobacteria, a group higher prokaryotes exhibiting multicellular social behaviors, possess predatory activity against plant pathogenic fungi and bacteria, giving them unique potential for application disease biocontrol. In this study, based on previously myxobacterial strain collection, strain, HM-E, broad-spectrum antifungal was screened. Through morphological observation, physiological biochemical characterization, multi-locus sequence analysis, identified as Cystobacter fuscus HM-E. C. HM-E not only significantly lysed V. hyphae but also inhibited its spore germination. Both cell-free fermentation filtrate volatile metabolites exhibited certain activity. Greenhouse pot assays showed that broth had efficacy 23.01% wilt, whereas solid agent formulated with white star flower chafer ( Protaetia brevitarsis ) frass achieved 70.90%, promoted seedling growth. Furthermore, crude extracts concentrated using macroporous resin acid precipitation no protein obtained ammonium sulfate disrupted cell wall membrane hyphae, induced intracellular reactive oxygen species (ROS) burst spores germ tube elongation. Enzyme substrate profile indicated several peptidases, lipases, glycoside hydrolases secreted might play important roles process are biocontrol factors. This study suggests novel agent, has great combating wilt.
