ABSTRACT Microbial communities of small freshwater bodies interact dynamically with environmental factors in unknown ways. Longitudinal sampling four ponds Knox County, Ohio, revealed relationships among antibiotic resistance genes (ARGs) and such as pH tannin concentrations. For each site, microbial were collected by filtration, metagenomes analyzed short-read sequencing. ARGs quantified using the ShortBRED pipeline to detect quantify hits a marker set derived from Comprehensive Antibiotic Resistance Database. The top 30 showed increased abundance at end growing season. two largest encode components Stenotrophomonas drug efflux pump powered proton-motive force ( smeABC ) mycobacterial global regulator that activates acid stress response mtrA ). prevalence modest correlation acidifying conditions (low high tannic acids). Acidity amplifies transmembrane difference component force, thus increasing cell’s energy available for function ARG expression. Association taxa was tested Kraken2/Bracken predictor profiles. profiles strongest dependence proportion Proteobacteria, whereas pond Cyanobacteria lowest counts. Efflux pumps SmeABC transcriptional activation MtrA incur large expenditures whose may be favored low external pH, where is maximal. IMPORTANCE Compared rivers lakes, ecosystems are understudied despite close contact agriculture recreation. Environmental microbes offer health benefits well hazards human contact. Small water act reservoirs drug-resistant organisms transfer (ARGs). Yet, public rarely aware potential exposure ARG-carrying recreational bodies. Little known about capacity remediate pollution which biochemical select against genes. This study analyzes how aquatic depend on levels.
