ABSTRACT Chayote ( Sechium edule ) has been traditionally used in Mexico to manage diabetes, owing its hypoglycemic, anti‐oxidant, and anti‐inflammatory properties. However, the specific effects mechanisms of chayote juice (SEJ) preventing pancreatic dysfunction remain underexplored. This study investigated protective underlying SEJ multiple low‐dose streptozotocin (MLD‐STZ)‐induced diabetic mouse model H 2 O ‐treated insulinoma cell line (MIN‐6). C57BL/6 mice were administered MLD‐STZ induce treated with SEJ. Diabetic parameters, islet morphology, protein expressions evaluated. MIN‐6 cells assess SEJ's at cellular level. Molecular docking studies performed elucidate interactions between phenolic compounds key proteins involved oxidative stress. significantly preserved structure insulin levels, reducing blood glucose levels. Mechanistically, suppressed expression apoptosis markers (caspase‐3, PARP, BAX) inflammatory (NF‐κB, iNOS, NLRP3) pancreas MIN6 cells, indicating protection β‐cells. Furthermore, upregulates endogenous anti‐oxidant (Nrf‐2, HO‐1), which are essential for mitigating stress‐induced stress diabetes circumstances. indicated that vicenin‐2 exhibit strong binding affinities KEAP‐1, Nrf‐2, HO‐1 proteins. demonstrates substantial through anti‐apoptotic, actions. These findings suggest could be developed as a functional food management.

Fibrosis is a hallmark of progressive kidney diseases. The overexpression profibrotic cytokine, namely transforming growth factor β (TGF-β) due to excessive inflammation and tissue damage, induces fibrosis. inhibition TGF-β signaling markedly limited in experimental disease models. Targeting signaling, therefore, offers prospective strategy for the management Presently, marketed drugs have numerous side effects, but plant-derived compounds are relatively safer more cost-effective. In this study, TGFβR-1 was targeted identify lead among flavonoids using various computational approaches, such as ADME/T (absorption, distribution, metabolism, excretion/toxicity) analysis, molecular docking, dynamics simulation. screening identified total 31 with drug-like properties compounds, 5 showed higher binding affinity TGFβR-1, Epicatechin, Fisetin, Luteolin ranking at top three (-13.58, -13.17, -10.50 kcal/mol, respectively), which comparable control drug linagliptin (-9.074 kcal/mol). also exhibited outstanding protein-ligand interactions. dynamic simulations revealed stable interaction these site TGFβR-1. These findings indicate that flavonoids, particularly Luteolin, may compete ligand-binding suggesting can be further evaluated development potential therapeutics against Further,

The prevalence of kidney-related diseases has been increasing and emerged globally as a leading cause mortality, especially in developing countries where they are considered neglected public health problem. Renal commonly progressive may irreversible loss organ function, eventually necessitating renal replacement therapy. Although different pharmaceuticals for the treatment these pathologies, uncertain effectiveness presence adverse effects have generated growing need development novel nephroprotective compounds. Because many medicinal herbs typically used Brazilian folk medicine to prevent cure kidney ailments, ethnomedicine play promising strategic role identifying adding new potential molecules pharmacological arsenal.
