Anemarrhena asphodeloides Bunge and Phellodendri Chinensis Cortex (AP) is a synergistic drug combination used to treat depression. However, the molecular mechanism underlying therapeuticeffects of AP requires further elucidation. To investigate potential in treatment depression its action. The model was established by chronic unpredictable mild stress (CUMS) treated with different doses AP. UPLC-Q-TOF-MS/MS network pharmacology methods were analyze composition targets Molecular docking, affinity ultrafiltration (AUF) cellular thermal shift assays (CETSA) techniques interaction between components key targets. therapeutic effects active ingredients explored LPS-induced cell models. ameliorated CUMS-induced despair behaviors, regulate M1 / M2 phenotypic balance microglia reduced neuroinflammation. Then, A total 10 brain identified mPFC CUMS rats. Based on these chemical compositions, PTGS2 found be at core regulatory through analysis. All had good PTGS2, among which Neomangiferin strongest binding capacity. can decrease activity, PGE2 production EP2 expression downstream, reduce inflammatory response. In BV2 cells, regulated imbalance M1/M2 microglia, release pro-inflammatory factors, decreased PEG2, EP2, cAMP, Epac1. improves benefits targeting signaling pathway, thereby promoting wider use

Behçet's disease (BD) is a multifaceted autoimmune disorder affecting multiple organ systems. Vascular complications, such as venous thromboembolism (VTE), are highly prevalent, around 50% of individuals diagnosed with BD. This study aimed to identify potential biomarkers for VTE in BD patients. Three microarray datasets (GSE209567, GSE48000, GSE19151) were retrieved analysis. Differentially expressed genes (DEGs) associated identified using the Limma package and weighted gene co-expression network analysis (WGCNA). Subsequently, diagnostic explored through protein-protein interaction (PPI) machine learning algorithms. A receiver operating characteristic (ROC) curve nomogram constructed evaluate performance Furthermore, immune cell infiltration analyses single-sample set enrichment (ssGSEA) performed investigate underlying mechanisms. Finally, efficacy listed drugs was assessed based on signature genes. The limma WGCNA 117 DEGs related PPI then selected 23 candidate hub Four (E2F1, GATA3, HDAC5, MSH2) by intersecting sets from three ROC construction demonstrated high accuracy these four (AUC: 0.816, 95% CI: 0.723-0.909). Immune revealed positive correlation between dysregulated cells ssGSEA provided insights into mechanisms development progression Additionally, therapeutic agent screening targeting employed systematic approach construct diagnosis dysregulation, suggesting macrophage involvement development. BD-induced VTE, agents identified.
