Tongmai Sini decoction (TSD), the classical prescriptions of traditional Chinese medicine, consisting three commonly used herbal medicines, has been widely applied for treatment myocardial infarction and heart failure. However, absorbed components their metabolism in vivo TSD still remain unknown. In this study, a reliable effective method using ultra-performance liquid chromatography coupled with hybrid quadrupole-Orbitrap mass spectrometry (UHPLC-Q-Exactive-MS/MS) was employed to identify prototype metabolites (rat plasma urine). Combined defect filtering (MDF), dynamic background subtraction (DBS), neutral loss (NLF) data-mining tools, total thirty-two major compounds were selected investigated vivo. As result, 82 identified or tentatively characterized vivo, including 41 alkaloids, 35 phenolic compounds, 6 saponins. Meanwhile, A 65 (40 alkaloids 25 compounds) identified. The metabolic reactions mainly hydrogenation, demethylation, hydroxylation, hydration, methylation, deoxylation, sulfation. These findings will be beneficial an in-depth understanding pharmacological mechanism pharmacodynamic substance basis TSD.
