The aim of this study was to investigate the effect curcumin nanocrystals (Cur-NCs) on ferroptosis in high-glucose (HG)-induced HK-2 cells and streptozotocin (STZ)-induced diabetic nephropathy model (DN) rats. purpose is determine whether Cur NCs can become a promising treatment option for diabetes by reducing ferroptosis. Cur-NCs were prepared using microfluidic technology studied dynamic light scattering transmission electron microscopy. treated with 30 mM HG create renal tubule damage cell model. Then, viability evaluated varying concentrations (0.23, 0.47, 0.94, 1.87, 3.75, 7.5, 15, μg/mL) Cell Counting Kit-8 (CCK-8). Furthermore, vivo experiments carried out roles STZ-induced DN results showed that greatly enhanced levels LDH, MDA, Iron, lipid ROS, apoptosis, NCOA4, TFR-1, while decreasing expression GSH, GPX4, SLC7A11, FTH-1. These effects induced could be attenuated Cur-NCs. also reduced HG-induced decrease viability, as well increase ROS however erastin inhibit their effects. inhibited This demonstrates significantly attenuate recovering implying may therapy DN.

Objective: Aconitine alkaloids, as the principal bioactive constituents of Fuzi, pose a significant challenge to its clinical application due their toxicity. This study aimed establish rapid, efficient, and stable method for quantifying monoester-type diester-type alkaloids in raw Fuzi using zeolitic imidazolate framework-8 (ZIF-8). The was subsequently applied pharmacokinetic studies rats, offering valuable insights into safe use Fuzi. Methods: Synthetic ZIF-8 employed microextraction adsorbent, with optimization extraction parameters such content, shaker speed, time, sodium ion concentration maximize enrichment efficiency. A dispersive solid-phase extraction-liquid chromatography-tandem mass spectrometry (d-SPE-LC-MS/MS) method, based on ZIF-8, developed validated performance. pharmacokinetics five aconitine were investigated, ensuring efficient analysis. Results: Under optimized conditions, d-SPE demonstrated robust alkaloids. strong linear relationship established aconitine, hypaconitine, mesaconitine, lappaconitine, benzoylaconitine within range 0.3125-1000 ng/mL, correlation coefficients exceeding 0.99. LC-MS/MS assay achieved detection limit low 0.104 ng/mL. Additionally, analysis revealed rapid absorption exhibiting Tmax 0.25 h. Conclusion: introduces novel d-SPE-LC-MS/MS plasma, facilitating These findings substantially contribute deeper understanding vivo

The escalating global prevalence of diabetes underscores the urgency addressing its treatment and associated complications. Paeoniflorin, a monoterpenoid glycoside compound, has garnered substantial attention in recent years owing to potential therapeutic efficacy management. Thus, this study aims systematically overview pharmacological effects, pharmacokinetics toxicity paeoniflorin diabetes. Plenty evidences have verified that improves complication through reducing blood sugar, enhancing insulin sensitivity, regulating gut microbiota autophagy, restoration mitochondrial function, regulation lipid metabolism, anti-inflammation, anti-oxidative stress, inhibition apoptosis, immune so on. Paeoniflorin possess characteristics rapid absorption, wide distribution, metabolism renal excretion. Meanwhile, studies suggested low acute toxicity, minimal subacute chronic no genotoxic or mutational toxic effects. In conclusion, paper elucidates application safety profile
