Depression is a serious global mental disorder. Numerous studies have found that depression may be closely related to decreased neurogenesis, neuroinflammation, neurotransmitter imbalance, and synaptic plasticity dysfunction. The pathogenesis of complex involves multiple signal transduction pathways molecular changes. PI3K/AKT pathway an essential signaling in neurons, which widely expressed emotion-related regions the brain. Therefore, play moderating role mood disorders. However, mechanism not been fully described. This review systematically summarized discussed its potential treatment depression. will help development antidepressants.

Introduction Alzheimer’s disease (AD) is a common neurodegenerative in the elderly. Its pathological features include: A lot of misfolding and abnormal aggregation amyloid protein (Aβ); Autophagy disorder, oxidative stress, neuroinflammation, phosphorylated tau synaptic dysfunction. Modern pharmacological studies have found that Paisinhin (PA) has beneficial effects on prevention treatment central nervous system diseases. This study aims to explore role mechanism PA AD through autophagy pathway, lay scientific foundation for development clinical strategies AD. Methods N2A APP cells were treated with different concentrations PA. Cell viability was detected by CCK-8 method. Western blotting expression levels proteins related production, Tau levels. flow transfecting Lc3 double fluorescent plasmid. After Aβ injected into hippocampus WT mice intraperitoneally, learning memory ability tested new object recognition, y maze water maze. The stress level kit. inflammatory factors RT-qpcr. Results not affected at PA, but PS1 significantly decreased 40μM. can obviously improve accumulation AD, some extent save inhibition CQ. Behavioral shown also impairments caused injections. In addition, vivo experiments, levels, inflammation salvage dysfunctions synapses. reduces total . Discussion Our provides first evidence improves Aβ-induced mice. effect appears be mediated promoting reducing stress. It may regulating inflammation, improving abnormally tau, salvaging damaged function, providing valuable insights potential applications

Gastrodia elata Blume (GE), a traditional Chinese medicine clinically employed to treat neurological disorders, demonstrates therapeutic efficacy supported by robust clinical evidence. Nowadays, conventional pharmacotherapies for conditions-such as cholinesterase inhibitors Alzheimer's or Ldopa Parkinson's-often provide limited symptom relief, exhibit side effects, and fail halt disease w, underscoring the need alternative strategies. The primary bioactive compounds of (GE) include gastrodin, p-hydroxybenzyl alcohol, Vanillyl Polysaccharides, β-sitosterol. Modern research has demonstrated that GE its active components neuropharmacological including neuron protection, reduction neurotoxicity, promotion nerve regeneration survival. For example, Gastrodin, exerts neuroprotection scavenging reactive oxygen species, suppressing pro-inflammatory cytokines, enhancing GABAergic transmission, thereby alleviating oxidative stress neuronal apoptosis. Vanillin, potentiates GABA receptor activity, inhibitory neurotransmission reducing seizure susceptibility.GE polysaccharides modulate gut-brain axis suppress microglial activation, mitigating neuroinflammation. Current studies primarily focus on ingredients treatment diseases such Parkinson's disease, epilepsy, convulsions, depression, schizophrenia, well learning memory, preventing treating cerebral ischemic injury. This review explores effects compounds, elucidates underlying mechanisms, suggests potential preventive strategies using herbal remedies.
