ABSTRACT Aims To evaluate the effects of Jiawei Kongsheng Zhenzhong Pill (JKZP) on rats with ischemic mild cognitive impairment (MCI) and investigate underlying mechanisms. Methods The components JKZP were analyzed using Q‐Orbitrap high‐resolution mass spectrometry (HRMS). MCI rat model was prepared through gradual bilateral common carotid artery occlusion (BCCAO). function, hippocampal pathological lesions, dendritic spine damage, synapse‐related, S100 calcium‐binding protein A10 (S100A10)/tissue‐type plasminogen activator (tPA)/brain‐derived neurotrophic factor (BDNF) pathway‐associated molecules alterations measured. Primary hippocampus neurons subjected to oxygen‐glucose deprivation/reperfusion (OGD/R) injury, JKZP‐containing serum utilized for treatment. Lentiviral‐infected constructed S100A10 knockdown RNAi technology whether exerted its anti‐MCI via S100A10/tPA/BDNF pathway. Results A total 64 major components, including β‐asarone, ferulic acid, loganin, senkyunolide H, cryptotanshinone, identified by HRMS technology. had a notable impact enhancing abilities MCI. reduced damage CA1 region neuron synaptic structure, reversed decrease in spines, increased expressions synapse‐associated proteins such as synaptophysin (SYN), growth‐associated 43 (GAP43), postsynaptic density 95 (PSD95). Furthermore, treatment dramatically ratio BDNF (proBDNF)/mature (mBDNF) activating S100A10/tPA, which confirmed primary vitro. Moreover, sh‐S100A10 tremendously mitigated inhibitory action OGD/R‐mediated synapse decreased activity tPA, thus improved downstream pathway targets’ ratio, proBDNF/mBDNF.
