The brown planthopper (BPH) is one of the major rice pests causing significant damage to production worldwide, due its high reproductive capacity and strong migratory ability. A series BPH-resistant varieties have been developed, but durability effective resistance constrained by evolution BPH virulence, requiring in-depth insights into mechanisms. In this paper, we used strand-specific lncRNA-seq characterize lncRNA regulatory mechanisms on response infestation. Overall, 4321 lncRNAs were identified, 60 which significantly upregulated in infestation, specifically differing between populations with variable virulence. Differential expression analysis qRT-PCR validation showed that these are involved regulation several defense pathways, including jasmonic acid signaling flavonoid biosynthesis, their distinct roles resistant susceptible varieties. Notably, like LNC_002533 found be negatively correlated suggesting a potential role modulating responses. contrast, LNC_001986 LNC_000397 positively genes glutathione metabolism, may associated enhanced resistance.
