Loss of lysosomal membrane integrity results in leakage hydrolases to the cytosol which might harm cell function and induce death. Destabilization lysosomes often precede apoptotic or necrotic death occur during both physiological pathological conditions. The weak base acridine orange readily enters cells accumulates acidic environment lysosomes. Vital staining with is a well-proven technique observe destabilization using fluorescence microscopy flow cytometry. These analyses are, however, time consuming only adapted for discrete points, make them unsuitable large-scale approaches. Therefore, we have developed time-saving, high-throughput microplate reader-based method follow real-time orange. This protocol can easily be adopted patient samples since number per sample low analysis short.

Although it has been known for decades that lysosomes are central degradation and recycling in the cell, their pivotal role as nutrient sensing signaling hubs recently become of interest. Since highly dynamic constant change regarding content intracellular position, fusion/fission events allow communication between organelles well cell-to-cell via exocytosis lysosomal release extracellular vesicles. Lysosomes also mediate different forms regulated cell death by permeabilization membrane to cytosol. In cancer cells, biogenesis autophagy increased support metabolism growth even under nutrient- oxygen-poor conditions. Tumor cells induce space promote invasion metastasis. However, due enhanced function, often more susceptible permeabilization, providing an alternative strategy death. This review summarizes current knowledge cancer-associated alterations structure function illustrates how vesicles affect disease progression. We focus on functional differences depending localization regulation transport, lastly provide insight new therapeutic strategies can exploit power lysosome improve treatment.
