The glycoside hydrolase 107 (GH107) family includes fucanase enzymes from only two bacterial phyla, Bacteroidota and Pseudomonadota. goal of this work was to explore the diversity putative related in organisms PVC superphylum (Planctomycetota, Verrucomicrobiota, Chlamydiota), order expand our knowledge fucoidan-degrading potential ecologically biotechnologically relevant group. Using hidden Markov model- peptide-based annotation tools, 26 GH107 homolog sequences were identified metagenome genome datasets. formed distinct clusters a phylogenetic analysis, one including members family. endo-acting fucoidan degrading activity confirmed an enzyme included most divergent cluster. fucanase, which probably originated uncultured planctomycete sampled subantarctic sediments, cloned expressed Escherichia coli. catalyzed rapid hydrolysis internal glycosidic bonds Macrocystis pyrifera, macroalgae species abundant at site. It active wide range temperatures (5–45 °C), salinities (9.5–861 mM NaCl), pH values (4.5–9), mainly producing sulfated α-(1,3)-linked fuco-oligosaccharides various lengths. represents promising source with biotechnological applications.
