Fungal chlamydospores are asexual spores formed by fungi under adverse conditions and could be used in biological pretreatment for biogas projects fed lignocellulosic substrates. In this study, Trichoderma viride (Tv) were as the agent to enhance methane yield of spent mushroom substrates (SMSs). Lignocellulosic composition, methanogenesis performance, anaerobic microbial communities investigated different Tv durations (0 h, 12 24 48 96 192 h). The results showed that optimal duration was cumulative reached 173.4 mL/gVS, which 16.8% higher than control. A longer h not conducive methanogenesis. Sequencing analysis directly proportional relative abundance at beginning digestion. When initial 50%, became absolute dominant fungus with an 97% fungal later stage correlation network among fungi, bacteria, archaea related 11 genera, through these taxa, affected 58% taxa whole network. Cost accounting has a net income 45.5 CNY/1000 kg SMS, is promising technology. This study provides important guidance use also promotes understanding
