Nanoplastics (NPs) cause serious contamination of drinking water and potential damage to human health. This study aimed investigate the effects NPs with different particle sizes concentrations on reproductive function male mice. In this study, free exposure was used expose BALB/C mice PS-NPs (20 nm, 200 1000 nm) at 0.1 mg/L, 1 5 mg/L for 4 months. The assessed after exposure, fecal blood samples were collected macrogenomics metabolomics. results showed that resulted in reduced testicular organ coefficients, decreased sperm quality, altered tissue structure, disturbed sex hormone levels, abnormal levels inflammatory factors oxidative stress. Furthermore, found NP affected alteration gut communities metabolic pathways related reproduction, such as Clostridium glutathione metabolism. Importantly, we an effect size function. future, more attention should be paid smaller NPs.

Viral infections of the reproductive tract and testis in male yaks, often resulting from natural mating under grazing conditions, can lead to infertility due Leydig cell (LC) apoptosis, immune activation, oxidative stress, reduced testosterone production. Spermidine (SPD), a potential therapeutic agent with antioxidant anti-aging properties, might alleviate oxidant response, virus infection caused by apoptosis. In this study, firstly testicular cells yak were induced Poly(I:C), pathogen-associated molecular pattern dsRNA virus, as pathogenic model at cellular level. Secondly, synthesis measured LC or without SPD culture medium. Finally, transcriptomic sequencing was utilized investigate mechanisms underlying protective effects SPD. These results suggested Poly(I:C) damaged function cells, significantly decreased concentration testosterone, while alleviated response then inhibited apoptosis restores production LCs. Transcriptomic analysis revealed that alleviates inflammation reducing damage through regulation several key gene expressions. findings suggest has ability mitigate Poly(I:C)-induced restore offering promising strategy protect enhance fertility after virus.

Objective The aim of this study is to use network pharmacology and data mining explore the role traditional Chinese medicine (TCM) in ischemic stroke (IS) intervention by ferroptosis regulation. results will provide reference for related research on IS. Methods ferroptosis-related targets were obtained from GeneCards, GeneCLiP3, FerrDdb databases, while IS sourced GeneCards DisGeNET databases. Venny was used identify associated with ferroptosis. A protein-protein interaction (PPI) analysis then conducted, machine learning screening validate these potential targets. that met specific criteria their compounds allowed us select TCMs. mechanistic conducted using DAVID database. PPI diagrams, target-compound target-compound-TCM diagrams constructed. Finally, molecular docking technology verify binding activities TCM core components identified In addition, properties, flavors, meridian tropism, therapeutic effects candidate TCMs analyzed statistically evaluated. Results total 706 obtained, 14 learning. Furthermore, 413 301 screened, prescriptions stable. primarily exhibited cold, warm, bitter taste, pungent liver meridian, heat-cleaning medicinal, tonify deficiency properties. Conclusions This investigated regulation TCM. We began investigating ferroptosis, we also relevant mechanism

Premature ovarian insufficiency (POI) is a disease with medical, psychological and reproductive implications, but its common therapies have limited efficacy likelihood of complications. This study delves into the therapeutic role human umbilical cord mesenchymal stem cell-derived extracellular vesicles (hUC-MSCs-EVs) in POI mice through insulin-like growth factor 1 (IGF-1)/nuclear E2 related 2 (Nrf2)/heme oxygenase-1 (HO-1)/autophagy pathway. hUC-MSCs were transfected lentiviral short hairpin RNA IGF-1 before EV extraction. Cyclophosphamide (CTX)-induced mouse models administrated hUC-MSCs-EVs. Mouse granulosa cells (GCs) induced CTX, then treated hUC-MSCs-EVs ML385. Ovarian histopathological changes observed, follicle number at all levels counted serum sex hormones evaluated, as well LC3II/I Beclin-1 expression. GCs subject to detection proliferation, deaths, oxidative stress, Nrf2 nuclear translocation. After CTX exposure, showed thinner layer ovary, reduced follicles levels, disturbed hormones, enhanced stress autophagy, downregulated IGF-1; whereas, upregulated improve function. carrying activated Nrf2/HO-1 signaling inhibit CTX-induced excessive autophagy GCs, this ameliorative effect was partially weakened by inhibiting signaling. inhibited improved function IGF-1/Nrf2/HO-1 activate IGF-1, which turn inhibits damage thus improving mice.

Ovarian aging and disease-related decline in fertility are challenging medical economic issues with an increasing prevalence. Polyamines a class of polycationic alkylamines widely distributed mammals. They small molecules essential for cell growth development. alleviate ovarian through various biological processes, including reproductive hormone synthesis, metabolism, programmed death, etc. However, abnormal increase polyamine levels can lead to damage promote the development disease. Therefore, polyamines have long been considered potential therapeutic targets disease, but their regulatory roles ovary deserve further investigation. This review discusses mechanisms by which ameliorate human disease different such as autophagy oxidative stress, develop safe effective targeted therapy strategies diseases.
