The Euphrates-Tigris catchment area is one of the major drainage basins in Middle East but climate change, overuse waters and subsequent salinization has brought this region to brink ecological devastation. aim study was investigate biodiversity epiphytic diatoms Iraq, for first time using a combination morphological metabarcoding methods. 47 samples were collected at six sampling sites along Rivers Tigris Euphrates northern Basra, Southern during summer 2019 (dry season) winter 2020 (rainy season). composition each sample compared with both microscopic-morphological analysis environmental resulted identification 284 infrageneric taxa 59 genera. For V4 18S marker gene used 1454 ASVs (Amplicon sequence variants) comprising 54 genera 108 species several belonging same taxa. non-metric multidimensional scaling (NMDS) plots morphology showed clear seasonal effect diatom community composition, while NMDS 18SV4 analyses did not show samples. incomplete taxonomic reference database studied turned out be limitation molecular approach. Nevertheless, methods increased detection assemblages laid foundation understanding their region.
