A significant number of silent biosynthetic gene clusters (BGCs) within the Burkholderia genome remain uncharacterized, representing a valuable opportunity for discovery new natural products. In this research, recombineering system ETh1h2e_yi23, which facilitates recombination in and was developed our previous study, used mining BGCs B. plantarii DSM9509. By using system, constitutive promoter precisely inserted into genome, resulting activation pla BGC, led to production lipopeptide named plantariitin A. distinctive characteristic is incorporation non-proteinogenic amino acid residue, i.e., amino-1,2,3,6-tetrahydro-2,6-dioxo-4-pyrimidinepropanoic (ATDPP), has not been identified other biological activity assay demonstrated that exhibits anti-inflammatory activity. This study further substantiates notion situ crucial strategy products genus Burkholderia. With increasing availability genomic data development bioinformatics tools, poised emerge as prominent source lipopeptides.

ABSTRACT Outer space is an extreme environment and the survival of many microorganisms after spaceflight well established. However, adaptations Bacillus subtilis to stress, particularly metabolism, are largely unknown. Here, we first performed a mission B. TD7 strain compared spaceflight‐exposed with wild‐type in terms their phenotype, biofilm formation secondary metabolism. The exhibited slower growth, different morphology decreased formation. Importantly, decline lipopeptide production was observed spaceflight. Multi‐omics approaches were used uncover molecular mechanisms underlying metabolism 997 differentially expressed genes (DEGs) found, involving TCA cycle, fatty acid degradation, amino biosynthesis quorum sensing systems. Further analysis 26 lipopeptide‐related DEGs further elucidated relationship between regulation. Our findings could contribute better understanding microbial adaptation mechanisms.

Antimicrobial resistance (AMR) has emerged as a significant crisis in global health. Due to their advantageous properties, antimicrobial peptides (AMPs) have garnered considerable attention potential alternative therapy address the AMR crisis. These might disrupt cell membranes or walls exhibit activity, modulate immune response promote recovery from diseases. In recent years, progress been made research of AMPs, alongside emergence new challenges. This review first systematically summarizes and critically discusses advancements understanding characteristics current landscapes well regulatory mechanisms action practical applications, particularly those reported approved within last 5 years. Additionally, principles, paths for identification, future trends AMPs are also analyzed following discussion advantages disadvantages comparison conventional antibiotics. Unlike prior literature this field, report summarized latest major discovery methods and, more importantly, emphasized applications by supporting various viewpoints using selected examples AMPs' real-life scenarios. Besides, some emerging hot topics including derived gut microbiota synergistic effects combating AMR, were profiled. All these indicate originality provide valuable references AMP discoveries applications.

Nonribosomal lipopeptides (NRLPs) are structurally complex natural products that play crucial ecological and biological roles. They also valuable sources lead structures for developing new pharmaceuticals. These compounds typically synthesized using a molecular assembly machinery known as nonribosomal peptide synthetases (NRPSs) or hybrid polyketide synthases-NRPSs. Unlike conventional NRPS, NRLPs characterized by starter module loads lipid chains substrate synthesis pathway supplies the necessary substrates during initiation stages. Unique critical determinants of activity NRLPs. Therefore, modifying these through combinatorial biosynthesis holds great promise unlocking their full therapeutic potential. Herein, we use term "Starter Unit" to refer initial modules lipoinitiation involved in chain process This Review provides comprehensive summary recent advances units offers insights into future directions further development.
