Microbial contamination is an important factor threatening the safety of Chinese medicine preparations, and microfluidic detection methods have demonstrated excellent advantages in application rapid bacterial detection. In our study, a novel optical biosensor was developed for sensitive Salmonella traditional on chip. Immune gold@platinum nanocatalysts (Au@PtNCs) were utilized specific labeling, while magnetic nano-beads (MNBs) with high-gradient field employed capture bacteria. The immune MNBs, Au@PtNCs, samples introduced into passive micromixer full mixing, resulting formation double-antibody sandwich structure due to antigen–antibody reactions. Subsequently, mixture flowed reaction cell, where MNBs-Salmonella-Au@PtNCs complex captured by field. After washing, hydrogen peroxide-tetramethylbenzidine substrate (H2O2-TMB) added, reacting Au@PtNCs peroxidase produce blue product. This entire process automated using portable device, concentration analyzed via phone application. simple has good specificity range 9 × 101–9 105 CFU/mL can detect concentrations as low 90 within 74 min. average recoveries spiked ranged from 76.8% 109.5%
