Accurate measurements of the molecular composition single cells will be necessary for understanding relationship between gene expression and function in diverse cell types. One most important phenotypes that differs is their size, which was recently shown to an determinant proteome populations similarly sized cells. We, therefore, sought test if effects size on protein concentrations were also evident single-cell proteomics data. Using relative a set reference proteins estimate cell's DNA-to-cell volume ratio, we found differences explain significant amount cell-to-cell variance two published data sets.

The mammalian mitochondrial genome (mtDNA) is multicopy and its copy number (mtCN) varies widely across tissues, in development disease. Here, we systematically catalog this variation by assaying mtCN 52 human tissues 952 donors (10,499 samples from the Genotype-Tissue Expression project) 20 murine using qPCR, capturing 50- 200-fold variation, respectively. We also estimate per cell 173 lines Cancer Cell Line Encyclopedia whole-genome sequencing data observe >50-fold variation. then leverage vast amount of genomics available for these repositories to credential our resource uncover mtDNA-related biology. Using already existing proteomics data, show that can be predicted TFAM, histone, ribosome protein abundance. integrate estimates with CRISPR gene dependency measurements find high are resistant loss GPX4, a glutathione phospholipid hydroperoxidase. Our captures should broadly useful research community.

Cells adopt a size that is optimal for their function, and pushing them beyond this limit can cause cell aging death by senescence or reduce proliferative potential. However, increasing genome copy number (ploidy), cells increase dramatically homeostatically maintain physiological properties such as biosynthesis rate. Recent studies investigating the relationship between rates of metabolism under normal, polyploid, pathological conditions are revealing new insights into how attain best function fitness tuning processes including transcription, translation, mitochondrial respiration. A frontier to connect single-cell scaling relationships with tissue whole-organism physiology, which promises reveal molecular evolutionary principles underlying astonishing diversity observed across tree life.
