Background: This study investigates the application of polyamidoamine (PAMAM) dendrimers as an innovative drug delivery approach for enhancing pharmacokinetic profile ursolic acid (UA), a pentacyclic triterpenoid with multifaceted therapeutic properties. UA, sourced from plants like Sanguisorba officinalis and Salvia officinalis, has been extensively studied its pharmacological characteristics, including anti-inflammatory, antioxidant, anti-diabetic properties, recognized in Traditional Chinese Medicine (TCM). The clinical utility UA is hampered by low bioavailability, which attributed to hydrophobic nature. To address this limitation, we explore use PAMAM dendrimers, known their potential. Methods: UA-PAMAM G0 were synthesized varying molar ratios. Characterization included size analysis, PDI, zeta potential determination. FTIR confirmed chemical structure. Male SD rats acclimatized administered control suspension UA-G0 dendrimer complex orally. Blood samples collected analysis. obtained IAEC approval. Results: complexes exhibited sizes based on ratios, 2:1 ratio showing significantly smaller dimensions. successful conjugation. In study, demonstrated higher plasma concentrations than alone, indicated increased Cmax AUC values. results suggest enhanced oral bioavailability complex. Conclusion: synthesis variations analysis revealed improved compared alone. These findings highlight compounds bridging gap between traditional herbal medicine modern strategies. Further research can broader applications such systems.
