Chemotherapy has been identified as a validated and critically important strategy for the treatment of cancer, but multidrug resistance serious side effects remain grand challenges effective cancer therapy. This highlights urgent need development alternative chemical entities that can modulate more than one biological target with high specificity multitargeted mechanism action in disease progression pathway. 1,2,3-Triazole hybrids have potential to act on dual/multiple targets cells simultaneously possess potent broad-spectrum activity against various cancers, including drug-resistant forms. Abstract Paracetamol‐based bis 1,2,3‐triazole analogues were synthesized involving copper‐catalyzed click chemistry protocol. The new compounds characterized by 1 H NMR, 13 C and mass spectral techniques. All the evaluated for their in vitro anti‐inflammatory properties using diclofenac as standard reference. compound 6b with chlorine fluorine functions displayed outstanding activity IC 50 values of 7.34 ± 0.45 µM 7.84 0.47 µM, respectively. Compound 6g methyl best 9.34 0.22 9.40 0.35 6i , 6k, 6f, 6a, 6e, 6h, 6j 6l exhibited promising respect to diclofenac. Using PyRx tool, molecular docking study against crystal structure COX‐2 proved binding efficacies notable interactions.
