Cardiac fibrosis is a common pathology in cardiovascular diseases which are reported as the leading cause of death globally. In recent decades, accumulating evidence has shown that biomechanical traits play important roles cardiac initiation, progression and treatment. this review, we summarize four main distinct (i.e., stretch, fluid shear stress, ECM microarchitecture, stiffness) categorize them into two different types static dynamic), mainly consulting unique characteristic heart. Moreover, also provide comprehensive overview effect on fibrosis, their transduction mechanisms, in-vitro engineered models targeting will aid identification prediction mechano-based therapeutic targets to ameliorate fibrosis.

Abstract Cells and tissue integrity is constantly challenged by the necessity to adapt respond mechanical loads. Among cellular components, nucleus possesses mechano-sensing mechanotransduction capabilities, yet molecular mechanisms involved remain poorly defined. We postulated that properties of chromatin its compartmentalization into condensates contribute nuclear adaptation external forces, while preserving integrity. By interrogating effects MLL4 loss-of-function in Kabuki Syndrome, we found balancing transcriptional Polycomb tunes responsiveness forces. showed acts as a mechano-sensor clustering through Prion-like domain, response was regulated context. Furthermore, activity instrumental withstand physical challenges nuclei experience during cell confinement migration their In Syndrome persistent rupture envelope triggers cGAS-STING activation, which leads programmed death. Ultimately, these results demonstrate critical role compartments play mechano-responses how they impact pathological conditions stimulating signaling.

The growing obesity epidemic necessitates increased research on adipocyte and adipose tissue function disease mechanisms that progress obesity. Historically, adipocytes were viewed simply as storage for excess energy. However, recent studies have demonstrated play a critical role in whole-body homeostasis, are involved cell communication, experience forces vivo, respond to mechanical stimuli. Changes the microenvironment can affect and, some cases, contribute disease. aim of this review is summarize current literature mechanobiology adipocytes. We reviewed over 100 papers how stress sensed by adipocyte, effects behavior, use culture scaffolds, particularly those with tunable stiffness, study properties, computational models. From our review, we conclude responsive stimuli, adipogenesis be dictated environment, measurement properties highly dependent testing methods, modeling practices many different approaches recapitulate complex behavior tissue. This intended aid future summarizing mechanobiology.
