Abstract Polyhydroxyalkanoate (PHA) as bioplastic is considered a replacement for conventional plastic due to its more beneficial properties. The ability of PHA biodegrade in shorter period major advantage. Different sewage water samples were collected from the Budha Nala near Maheru regions Punjab. PHA-producing bacteria isolated using minimal salt media supplemented with Nile blue. Further screening was carried out Sudan Black B stain and red stain. positive isolates characterized gram reaction, motility, biochemical tests. individual later screened maximum accumulation glucose. extracted FTIR, XRD, SEM, UV spectroscopy, NMR, TGA. Twenty-six different on Upon stain, nineteen showed black granules orange fluorescence bodies under 100X magnification that confirmed polyhydroxyalkanoates. tests partially belonging Bacillus genus. All produced granular form, however, isolate P-3 production 0.068 g/L. FTIR XRD chemical crystallinity studies spectroscopy by analyzing absorption spectra at 235 nm standard crotonic acid sulfuric conversion acid. P-3, paranthracis RSKS-3 first reported bacterium produce necessary optimize efficiency yield.
