Abstract Opioid addiction is a complicated and highly heritable brain disease. Dysfunction of dopaminergic signaling involved in the pathogenesis addictive disorders. Encoding synthetase dopamine, tyrosine hydroxylase (TH) gene has long been an interesting candidate genetic association studies for opioid addiction. However, underlying mechanism risk variants remains unknown. In present study, we first analyzed between TH susceptibility traits heroin 801 patients with 930 healthy controls. The methylation level promoter region was detected compared control groups. To reveal potential addiction, correlation SNPs as well expression were examined. Our results demonstrated that SNP rs6356 associated CpG TH_ 15 hypermethylated group group. Notably, allele-specific correlated hippocampus nucleus accumbens, but not 15. findings suggest eQTL by potentially affecting mesocorticolimbic dopamine system regions including accumbens.
