Abstract DNA methylation, a pivotal epigenetic modification, plays critical role in regulating gene expression. Understanding its dynamics with age is essential for elucidating ageing processes and developing biomarkers. This short note presents theoretical framework modelling methylation using fractional calculus approach, extending standard exponential models to accommodate the complex, heterogeneous nature of changes over time. The fractional-calculus representation process, described by fractional-order differential equation solution based on Mittag-Leffler function, not only provides better results sense R 2 , RMSE, MSE criteria but also offers more general model, where model one special case.
