Abstract Memories are encoded in neural ensembles during learning 1–6 and stabilized by post-learning reactivation 7–17 . Integrating recent experiences into existing memories ensures that contain the most recently available information, but how brain accomplishes this critical process remains unclear. Here we show mice, a strong aversive experience drives offline ensemble of not only memory also neutral formed 2 days before, linking fear to previous memory. Fear specifically links retrospectively, prospectively, across days. Consistent with studies, find is reactivated period after learning. However, increases co-reactivation period. Ensemble occurs more wake than sleep. Finally, expression context associated shared between memories. Collectively, these results demonstrate mechanism which integrated

Cognitive processes such as action planning and decision-making require the integration of multiple sensory modalities in response to temporal cues, yet underlying mechanism is not fully understood. Sleep has a crucial role for memory consolidation promoting cognitive flexibility. Our aim identify sleep integrating different enhance flexibility task execution while identifying specific brain regions that mediate this process. We have designed "Auditory-Gated Patience-to-Action" Task which mice should process auditory signals before well analyzing visual inputs feedback their action. Mice could learn rule apply it only after sleeping period keep performance constant across sessions. c-fos positive cells showed involvement prelimbic cortex (PrL) during execution. Chemo-genetic inhibition verified PrL required proper signal timing. These findings emphasize cortical activity are keys adapting modalities.

ABSTRACT Idling brain activity has been proposed to facilitate inference, insight, and innovative problem-solving 1-4 . However, it remains unclear how when the subconscious can create novel ideas. Here, we show coordinated roles of non-rapid eye movement (NREM) rapid-eye (REM) sleep in weaving inferential information from learned information. In a transitive inference paradigm, mice gained one day, but not shortly, after complete training. Inhibiting neuronal computations anterior cingulate cortex (ACC) during post-learning either NREM or REM sleep, wakefulness, disrupted without affecting knowledge. vivo Ca 2+ imaging showed gradual development inference-related representation, peaking at post-training sleep. Neurons representing co-reactivated significantly while their co-reactivation with neurons was higher Furthermore, insufficient learning, artificial activation medial entorhinal cortex-ACC dialogue only created These findings suggest that organizes scattered knowledge hierarchy, computes organized hierarchy. Collectively, our study provides mechanistic insight on coordination knowledge, thus highlighting power idling cognitive flexibility.

However, it still unclear what kind information processing occurs brain during effects processing-related events are transferred from wakefulness sleep. Research suggests that has a positive impact function. whether specific stages, such as NREM REM, exclusively dedicated certain types memory, semantic or event memory. When evaluating research approaches discussed above, can be concluded important note limitations studying challenges conducting this state. Despite spending third lives asleep, benefits remains limited. It fact investigating phenomenon sleep, help us understand learning, consciousness, well function neurophysiology.
