Gut microbiota dysbiosis is associated with multiple sclerosis (MS), but the causal relationship between specific gut bacteria and MS pathogenesis remains poorly understood. Therefore, we profiled stool microbiome of people (PwMS) healthy controls (HC) using shotgun metagenomic sequencing. PwMS showed a distinct compared to HC, Prevotella copri (PC) Blautia species as drivers microbial communities in HC PwMS, respectively. Administration MS-driving (Blautia wexlerae; BW) mice resulted increased levels inflammatory markers altered capacity induce proinflammatory cytokines. Utilizing experimental autoimmune encephalomyelitis (EAE), an animal model MS, identified lower Bifidobacterium Akkermansia ratio hallmark disease. BW-administered also pre-EAE induction which correlated disease severity post-EAE induction. The importance at level, adolescentis muciniphila (BA:AM), was validated our cohort large International Multiple Sclerosis Microbiome Study. Thus, findings highlight BA:AM potential marker opening avenues for microbiome-based diagnosis, prognosis, therapy MS.
