Radial neuronal migration is a key neurodevelopmental event for proper cortical laminar organization. The multipolar-to-bipolar transition, critical step in establishing polarity during radial migration, occurs the subplate/intermediate zone (SP/IZ), distinct region of embryonic cerebral cortex. It has been known that extracellular matrix (ECM) molecules are enriched SP/IZ. However, molecular constitution and functions ECM formed this remain poorly understood. ABSTRACT INTRODUCTION Alzheimer’s disease (AD) lacks a less invasive and early detectable biomarker. Here, we investigated the biomarker potential of miR-501-3p miR-502-3p using different AD sources. METHODS MiR-501-3p expressions were evaluated in CSF exosomes, serum familial sporadic fibroblasts B-lymphocytes by qRT-PCR analysis. Further, analyzed APP, Tau cells media exosomes. RESULTS significantly upregulated exosomes relative to controls. MiRNA levels high accordance with amyloid plaque NFT density multiple brain regions. Similarly, both miRNAs elevated MCI compared MiR-502-3p expression was fAD sAD B-lymphocytes. Finally, intracellularly secreted extracellularly response APP pathology. The formation of precise numbers neuronal connections, known as synapses, is crucial for brain function. Therefore, synaptogenesis mechanisms have been one the main focuses neuroscience. Immunohistochemistry a common tool visualizing synapses. Thus, quantifying synapses from light microscopy images enables screening impacts experimental manipulations on synapse development. Despite its utility, this approach paired with low throughput analysis methods that are challenging to learn and results variable between experimenters, especially when analyzing noisy tissue. We developed an open-source ImageJ-based software, SynBot, address these technical bottlenecks by automating analysis. SynBot incorporates advanced algorithms ilastik SynQuant accurate thresholding synaptic puncta identification, code can easily be modified users. use software will allow rapid reproducible phenotypes in healthy diseased nervous systems.

Abstract In conditions such as neurodegenerative diseases, posttraumatic stress disorder (PTSD), addiction and spinal cord injuries, restricted synaptic plasticity hinders the formation of new neuronal connections, preventing compensation treatment adverse behaviors. Perineuronal nets (PNNs) significantly restrict by inhibiting synapse formation. The digestion PNNs has been associated with short-term cognitive improvements reduced long-term memory, offering potential therapeutic benefits in PTSD. This study investigates correlation between fear memory processes extracellular matrix (ECM) mutant mice, particularly focusing on amygdala-medial prefrontal cortex (mPFC) circuit, which is crucial for generation maintenance. Fear conditioning was conducted mice lacking four key ECM-molecules: brevican, neurocan, tenascin-C tenascin-R (4x KO). These exhibited severe impairments consolidation, evident their inability to retrieve previously learned memories, coupled PNN density disturbed integrity along PNNs. Additionally, changes neural activity basolateral amygdala (BL) reductions VGAT + puncta amygdala-mPFC circuit were observed. contrast, tenascin single KOs showed intact behavior compared control groups.
