Abstract
The
β-secretase
BACE1
has
become
a
prime
target
in
Alzheimer’s
disease
(AD)
therapy,
because
it
drives
the
production
of
pathogenic
amyloid
β
peptides.
However,
clinical
trials
with
BACE1-targeting
drugs
were
halted
due
to
adverse
effects
on
cognitive
performance.
We
propose
here
that
impairment
by
inhibitors
may
be
corollary
higher
function
related
proper
sleep
regulation.
To
address
non-enzymatic
ion
channels
likely
involved
sleep–wake
cycle,
we
analyzed
patterns
both
BACE1-KO
mice
and
newly
generated
transgenic
line
expressing
proteolysis-deficient
variant
(BACE1-KI).
found
BACE1-KI
displayed
common
distinct
disturbances.
Compared
their
respective
wild-type
littermates,
mutant
lines
slept
less
during
light
phase
(when
they
preferentially
rest).
Furthermore,
transition
rates
between
wake
states
altered,
as
spindles
EEG
power
spectra
gamma
range.
Thus,
better
understanding
how
interferes
sleep-modulated
behaviors
is
needed
if
BACE1-targeted
are
resume.

used

traditional
folk
medicines
for
treatment

ailments.
Modern
research
revealed
that
aspera
possesses
pharmacological
effects,
including
cardiocerebrovascular
protection,
immune
regulation,
antioxidation,
anti-aging.
Furthermore,
neuroprotective
effects
confirmed
numerous
scientific
studies.

on

mechanisms
prevention

diseases,
as
well
potential
