Due to the radiosensitivity and deep anatomical location of nasopharyngeal carcinoma (NPC), radiotherapy serves as cornerstone standardized treatment for this malignancy. Beyond its cytotoxic effects, can serve an immunological adjuvant by inducing immunogenic cell death (ICD). Dendritic cells (DCs), potent antigen-presenting cells, play a critical role in tumor immunotherapy, but their exact ICD process NPC remains unclear. The effects high-dose radiation (≥2 Gy) on DCs type immune response it elicits have not been fully elucidated. An vitro study was conducted assess whether 5-8F induced could regulate DCs. Specifically, maturation capacity were evaluated following co-culture with exposed radiation. High-dose found induce evidenced increased pro-inflammatory factor levels reduced anti-inflammatory culture supernatant. Co-culture 15 minutes significantly enhanced expression surface molecules DCs, promoting sensitization. radiation-induced apoptosis is form ICD, which plays important regulating DC function. These findings provide insight into immunomodulatory potential enhance immunotherapy through activation.

Arsenic trioxide (As2O3) has long been utilized in traditional Chinese medicine due to its therapeutic properties. While it exhibits potent anticancer activity, clinical application is hindered by systemic toxicity and limited tissue specificity. In this study, an advanced approach developed using arsenic prodrug-loaded bimetallic sulfide MnZnSX nanorods (As-MnZnSX NRs) enhance both the efficacy safety of As2O3 hepatocellular carcinoma treatment. These are engineered release Mn2+ H2S within tumor microenvironment, facilitating binary-cooperative activation cGAS-STING pathway. This dual mechanism enhances immune responses while converting prodrug into cytotoxic form, AsIII. The results demonstrate that amplifies pathway inducing TBK1 phosphorylation IRF3 activation, leading dendritic cell maturation improved antigen cross-presentation. Simultaneously, promotes conversion collectively driving binary stimulation strategy significantly augments antitumor integrating modulation with targeted effects. Furthermore, enable vivo MRI, allowing real-time monitoring treatment progression. study represents a substantial advancement liver cancer therapy chemoimmunotherapy diagnostic imaging, thereby improving precision minimizing toxicity.
