ABSTRACT Amphibians are extremely sensitive to environmental changes, and their gut microbiome may have different responses changes. Here, metagenomic sequencing was used investigate the intestinal microbiota of Asiatic toad ( Bufo gargarizans ) from three habitats (city areas, transition wild areas) Sichuan Province, China. The results showed that Proteobacteria, Firmicutes, Fusobacteria were main bacteria in B. . There significant differences composition function among samples habitats. Enterobacteriaceae changes occupied a high relative abundance city especially for Citrobacter Especially, antibiotic resistance genes (ARGs) virulence factors (VFs) significantly increased areas. We performed de novo assembly metagenome‐assembled genomes (MAGs). In total, 322 nonredundant MAGs reconstructed, 304 which might be potential novel Among 13 species‐level genome bins (SGBs) belonging Enterobacteriaceae, one portucalensis annotated most types ARGs VFs. Phylogenetic functional analyses assembled C. MAG public data carried out, suggesting it play role diseases amphibians. Our study revealed across suggests amphibian could serve as indicators reflect human activities. reconstructed expanded our understanding amphibians, substantial reservoir resources.

Background: To explore the plasmid characteristics and transfer mechanisms of an extensive drug resistant (XDR) clinical isolate, Citrobacter portucalensis L2724hy, co-producing bla SFO-1 , NDM-1 KPC-2 . Methods: Species confirmation L2724hy was achieved through 16S rRNA sequencing Average Nucleotide Identity (ANI) analysis. Antimicrobial susceptibility testing (AST) employed agar dilution micro broth methods. Identification resistance genes carried out by PCR whole-genome (WGS). Essential gene locations were verified S1 nuclease pulsed-field gel electrophoresis (S1-PFGE) southern hybridization experiments. Subsequent WGS data analysis delved into plasmids. Results: The strain as drug-resistant portucalensis, to almost all antibiotics tested except polymyxin B tigecycline, sequencing, ANI AST results. subsequent revealed carrying on plasmids various sizes. uncommon ESBL coexists with fosA3 IncFII plasmid, featuring genetic environment IS 26-fosA3 -IS 26-ampR-bla 26 found IncX3 coexisting SHV-12 displaying sequence 5 3000 -Tn 2-bla - ble-trpF-dsbD-cutA-gros-groL lacking Aa125 is located unclassified exhibiting Tn 2-tnpR Kpn27-bla Kpn6-korC Conjugation assays confirmed transferability both Conclusion: We discovered coexistence in C. for first time, delving mechanisms. Our finding highlights importance vigilant monitoring drug-resistance insertion elements strains. Keywords: spp XDR, SFO NDM KPC
