Abstract Receptor tyrosine kinases (RTKs), a category of transmembrane receptors, have gained significant clinical attention in oncology due to their central role cancer pathogenesis. Genetic alterations, including mutations, amplifications, and overexpression certain RTKs, are critical creating environments conducive tumor development. Following discovery, extensive research has revealed how RTK dysregulation contributes oncogenesis, with many subtypes showing dependency on aberrant signaling for proliferation, survival progression. These findings paved the way targeted therapies that aim inhibit crucial biological pathways cancer. As result, RTKs emerged as primary targets anticancer therapeutic Over past two decades, this led synthesis validation numerous small molecule kinase inhibitors (TKIs), now effectively utilized treating various types. In manuscript we provide comprehensive understanding context We explored alterations specific receptors across different malignancies, special dedicated examination current inhibitors, highlighting potential therapies. By integrating latest evidence, seek elucidate pivotal biology efficacy inhibition promising treatment outcomes.

Abstract Liver cancer represents a major global health concern, with projections indicating that the number of new cases could surpass 1 million annually by 2025. Hepatocellular carcinoma (HCC) constitutes around 90% liver and is primarily linked to factors incluidng aflatoxin, hepatitis B (HBV) C (HCV), metabolic disorders. There are no obvious symptoms in early stage HCC, which often leads delays diagnosis. Therefore, HCC patients usually present tumors advanced incurable stages. Several signaling pathways dis-regulated cause uncontrolled cell propagation, metastasis, recurrence HCC. Beyond frequently altered therapeutically targeted receptor tyrosine kinase (RTK) involved differentiation, telomere regulation, epigenetic modification stress response also provide therapeutic potential. Abstract Diffuse large B-cell lymphoma (DLBC) is the most common subtype of non-Hodgkin lymphoma, characterized by its aggressive nature and poor prognosis in advanced stages. Despite advances treatment, molecular mechanisms driving DLBC progression remain incompletely understood, necessitating identification novel biomarkers for diagnosis prognosis. In this study, we analyzed two publicly available datasets (GSE32018 GSE56315) from Gene Expression Omnibus database (GEO) to identify overlapping differentially expressed genes (DEGs). Later on, a comprehensive silico vitro methodology was adopted decipher role DEGs DLBC. analysis GSE32018 GSE56315 identified five gene: SP3, CSNK1A1, STYX, SIRT5, MGEA5. validation using GEPIA2 confirmed upregulation downregulation MGEA5 tissues compared normal controls. Furthermore, mutational revealed that CSNK1A1 only gene among these exhibit mutations, with 2.7% mutation frequency patients. Methylation highlighted negative correlation between methylation levels mRNA expression, while survival high STYX expression as significantly associated poorer overall Functional assays demonstrated knockdown U2932 cells led reduced cell proliferation, colony formation, enhanced wound healing, indicating STYX’s pivotal migration. Additionally, enrichment involvement key biological processes, including intracellular trafficking myeloid progenitor differentiation. These findings emphasize potential therapeutic targets DLBC, particularly highlighting promising prognostic marker target intervention.
