Abstract Ulcerative colitis (UC) is a chronic, relapsing inflammatory disorder of the gastrointestinal tract. Programmed cell death (PCD), such as PANoptosis and autophagy, involved in various inflammation- immune-related diseases. This study aimed to examine molecular signature associated immune infiltration PANoptosis- autophagy-related differentially expressed genes (DEGs) UC. Ten PANoptosis-related hub DEGs, including PDGFRB, TIMP1, MMP2, CD44, TIMP2, TGFB2, IL6, TIMP3, IL1B, HGF, four CCL2, TGFB1, PPARG, CXCR4, were identified. These with chemotaxis, wound healing positive regulation MAPK cascade. Immune analysis revealed UC patients exhibited higher immunocytes these closely related infiltrations. Through machine learning algorithms, five key candidate genes, showed good diagnostic performance distinguishing from healthy individuals. Furthermore, highly fibroblasts (IAFs) by single-cell sequencing analysis. The expression levels significant differences colon mucosa patients. Our findings indicated that autophagy or crosstalk between them might be implicated dysregulation via regulating specific cells IAFs interacting signals chemotaxis signaling pathways.
