Abstract Background Cuproptosis, as a copper-induced mitochondrial cell death, has attracted extensive attention recently, especially in cancer. Although some key regulatory genes have been identified cuproptosis, the related lncRNAs not further studied. Background: Long noncoding RNAs (lncRNAs) as critical molecules play an essential role in the development of cancers. In colorectal cancer (CRC), various lncRNAs are related to cell proliferation, apoptosis, migration, and invasion. LncRNA prostate cancer-associated transcript 1 (PCAT-1), oncogenic factor, is a diagnostic biomarker that regulates invasion, apoptosis. Methods: This study evaluated relationship between PCAT-1, CRC occurrence, pathological features Iranian patients. The studied samples included 100 tumor tissues adjacent healthy were extracted from cancerous noncancerous synthesize complementary DNA. expression level PCAT-1 was assessed using real-time PCR method, data analysis SPSS software. Results: this study, tissue significantly increased patients, studies patients had no significant with profile. Conclusion: Our results suggested high resulted occurrence which can be considered CRC.

Background: Cellular senescence, a novel hallmark of cancer, is associated with patient outcomes and tumor immunotherapy. However, at present, there no systematic study on the use cellular senescence-related long non-coding RNAs (CSR-lncRNAs) to predict survival in patients osteosarcoma. In this study, we aimed identify CSR-lncRNAs signature evaluate its potential as prognostic marker predictive tool for immune response Methods: We downloaded cohort osteosarcoma from The Cancer Genome Atlas (TCGA) Gene Expression Omnibus (GEO) databases. performed differential expression co-expression analyses CSR-lncRNAs. univariate multivariate Cox regression along random forest algorithm lncRNAs significantly correlated senescence. Subsequently, assessed models using curves, receiver operating characteristic nomograms, C-index, decision curve analysis. Based model, were divided into two groups according their risk scores. Then, Ontology Kyoto Encyclopedia Genes Genomes analyses, compared clinical characteristics uncover functional differences. further conducted infiltration estimation stromal cells malignant tissues data (ESTIMATE), cell-type identification by estimating relative subsets rna transcripts (CIBERSORT), single-sample gene set enrichment analysis groups. also evaluated target genes checkpoint inhibitors (ICIs). Results: identified six that senescence accordingly established lncRNA incorporating these lncRNAs. nomogram indicated model was an independent factor could This demonstrated high accuracy upon validation. Further revealed low-risk group exhibited better enhanced infiltration. Conclusions: six-CSR-lncRNA effectively predicted might have improved

Abstract Background Colorectal cancer (CC) is one of the major contributors to tumor-related death worldwide, and its main cause distant metastasis. Dysregulation long non-coding RNA (lncRNA) LINC01605 has been implicated in CC. However, role metastasis CC remains elusive. The goal study uncover biological function molecular mechanism Methods differentially expressed lncRNAs were first screened from GSE97300, GSE84983, GSE110715, GSE70880, GSE75970 microarrays. correlation between expression clinical phenotypes enrolled patients (n = 134) was subsequently analyzed. upstream downstream regulatory mechanisms identified through bioinformatics RNA-seq analyses. Finally, effects related factors on cell growth confirmed functional validation experiments. Results LINC01605, significantly highly CC, a prognostic factor for with Functional experiments revealed that knockdown inhibited proliferatory metastatic potential cells vitro vivo. Moreover, regulated by SMYD2-EP300-mediated modifications histone H3K4me3 as well H3K27ac. found bind METTL3 promote m6A modification SPTBN2 mRNA, thereby facilitating translation SPTBN2. Conclusions Overexpression H3K27ac modifications, bound protein leading development

There is growing evidence that long non-coding RNAs (lncRNAs) are significant contributors to the epigenetic mechanisms implicated in emergence, progression and metastasis of colorectal cancer (CRC), but many remain underexplored. A novel lncRNA LOC105369504, was identified be a potential functional by microarray analysis. In CRC, expression LOC105369504 markedly decreased resulted distinct variations proliferation, invasion, migration epithelial-mesenchymal transition (EMT) vivo vitro. This study showed bound protein paraspeckles compound 1 (PSPC1) directly regulated its stability using ubiquitin-proteasome pathway CRC cells. The suppression could reversed through PSPC1 overexpression. This under-regulated as lncRNA, exerted tumor suppressive activity suppress proliferation together with cells regulation PSPC1. These results offer new perspectives on effect CRC.
