ABSTRACT Benzothiazole alkaloids are a class of rare heterocyclic with unique structures and exhibit wide range biological activities. So, the aim this work is to investigate structural diversity‐guided N‐S derivatives based on natural benzothiazole as potential cytotoxic agents. Three series novel derivatives, including 22 compounds, were designed synthesized using pharmacophore hybridization, their in vitro activities against Huh‐7 A875 fully evaluated. The results indicated that some these had significantly good two tested cell lines compared positive control 5‐fluorouracil, other compounds 3f–3i displayed selectivity between lines, which might be used promising lead molecule for discovery benzothiazole‐type
