Triple-negative breast cancer (TNBC) is a subtype of that has worse prognosis than the other subtypes because its high recurrence and metastasis rates. The objective this study to identify regulatory factors are associated with disease-free survival (DFS) TNBC potential biomarkers for treatment. We obtained GSE97342 dataset from Gene Expression Omnibus website conducted weighted gene co-expression network analysis (WGCNA) modules DFS TNBC. Subsequently, biological functions were elucidated through Ontology (GO) Kyoto Encyclopedia Genes Genomes (KEGG) pathway analyses. Cross-checking Human Transcription Factor Database facilitated selection hub transcription univariate Cox regression overlapping factors. Utilizing bioinformatics analysis, we assessed prognostic significance these factors, investigated their target genes, explored associations tumor immune cells in Finally, expression levels validated by immunohistochemical staining, quantitative reverse polymerase chain reaction (qRT-PCR), Western blotting. Through WGCNA identified three correlated GO KEGG analyses genes within modules. Background: Non-alcoholic fatty liver disease is related to lipid accumulation and inflammation. Considering the role of lipin-1 lipin-2 in fat homeostasis inflammation, this study aimed explore effect galbanic acid (Gal) resveratrol (RSV) on alterations gene expression levels protein abundance HepG2 cells lipid-enriched with palmitate (Pal). Materials Methods: were subjected different amounts Gal RSV for 24 hours presence Pal induce accumulation. The RT-PCR method was employed assess genes, while evaluated by western blot analysis. Lipid determined qualitatively semi-quantitatively using oil-red staining technique. Results: treatment increased ( P < 0.05). In contrast, groups treated did not show a substantial variance two genes > Gal/RSV, intensity bands higher compared group 0.01); however, significantly Conclusion: Gal, coumarin compound, Pal. Consequently, research suggests gal as novel strategy regulating cells.
